Are the same Internal Sales associates assigned to these Regional Sales Managers?  If so you could use the Team in CRM to associate these ISAs with the appropriate managers.  If there is not a direct team relationship you could add a field to the Contact/Lead for the ISA.  Make it a look up field to the User and then you have a hook.  You can create views and kick off workflows based on that user instead of the Owner.
